Professional Summary

Bala Addepalli is an Analytical Biochemist with broad research interests in analytical chemistry, biochemistry, molecular biology, mass spectrometry (MS)-based characterization and structural determination of proteins and nucleic acids. He described the type and nature of viral nucleic acids along with their protein coding potential during his PhD studies.  His postdoctoral studies with Prof. Arthur Hunt had identified the existence of novel RNA processing endonuclease in plant polyadenylation apparatus. His studies also unraveled the molecular characteristics of poly(A) polymerases and interactions with other key polyadenylation subunit factors. Later he worked with Prof. Patrick Limbach to develop mass spectrometry (MS)- based approaches through better sample preparation, improved detection and quantification of mass-silent nucleoside modification, pseudouridine, and other post-transcriptional modifications in RNA.

He was inducted into the faculty as Assistant Professor-Research at Department of Chemistry in August 2013 and was promoted to Associate Professor-Research in August 2018. The current research program includes development of novel ribonucleases for RNA modification mapping, improved characterization of nucleoside modifications (both enzymatic and xenobiotic stress-induced) and understanding the effect of xenobiotic (organic pollutants, metals and radiation)-induced changes to RNA and its post-transcriptional modificationprofiles leading to disease development. His research has been develping assays to probe the impact of xenobiotic stress on cellular translation using a variety of prokaryotic and eukaryotic model cell systems.

Research and Practice Interests

The research interests of Bala Addepalli revolve around the functional characterization of enzymatic and stress-induced ribonucleoside modifications (extra chemical groups attached to canonical, A, G, C, U) found in RNA. These studied aim at improved analytical method development and functional assays. Major objectives include
(1) development of novel biochemical tools, such as ribonucleases, to map (ability to locate) the modifications on to the RNA sequence;
(2) understanding the cellular and biological significance of nucleoside modifications (natural and xenobiotic);
(3)  elucidation of the effects of stress (including organic pollutants and radiation) on nucleoside modifications (through liquid chromatography mass spectrometry) and mRNA translation, and
(4) development of approaches that mitigate the stress-induced damage to the modified RNA.

03-2015 -04-2015 Development of novel ribonucleases for sequencing chemical modifications in RNA Discovery and quantification of chemical modifications in cellular RNA requires concerted effort through a combination of biochemical and analytical technologies. We demonstrate the utilization of novel nucleoside specific ribonucleases to sequence the location of chemical modifications in RNA. The RNA cleavage properties, nucleoside specificity and the substrate recognition behavior towards modified nucleosides are systematically characterized by the state-of the art liquid chromatography coupled with mass spectrometry. A grant proposal resubmitted to  UC Technology Acclerator for commercialization on "Novel Ribonucleases for Sequencing Chemical Modifications in RNA".
Abstract:
Reliable and unambiguous determination of the sequence location of a chemical modification is a critical requirement for evaluating the role of >150 such modifications found in natural RNA. We have recently made a disclosure about the development of enhanced molecular tools, viz., cytidine-specific and uridine-specific ribonucleases that aid in this process. These novel enzymes complement and enhance the utility of currently available ribonucleases such as RNase T1 (G-specific), RNase U2 (A/G-specific) and RNase A (C/U-specific). Based on the feedback from our previous application, we have revised our business model to focus on the development and distribution of these reagents. In this revised submission, we are seeking support that will enable us to investigate new end-use demonstrations of our reagents and support for an in-depth evaluation that will clarify the specific market(s) for our products. This funding will accelerate our entry into the billion-dollar “RNA analysis” field associated with pharmaceutical and biotechnological research.

"Development of novel research tools and a database for mapping human mitochondrial tRNA modifications by mass spectrometry-Phase I" for NIH funding      ,

An application submitted to the Ohio Cancer Research in March 2018 to investigate Role of RNA modifications in melanoma progression and invasion was not funded.,

 A collaborative research proposal with Prof. Romerio Fabio, University of Maryland, was submitted to NIH for funding support consideration. The title of the project is "Sustained HIV remission via sequence-specific epigenetic silencing of latent proviruses", with an expected duration of 5 years. My role in this project is a collaborator to characterize the RNA modifications of HIV-antisense RNA. ,

Analytical Core proposal was submitted as part of Superfund Research Project entitled, "Novel Strategies for Sensing, Investigating, and Remediating Exposure to Superfund Toxicants Affecting Reproduction" was submitted to NIH for funding support consideration.,

 A collaborative grant proposal has been submitted with Prof. Ken Pang at Department of Pediatrics, University of Melbourne for funding support to the Australian Government National Health and Medical Research Council Idea Grants 2019 program. The goal of the subaward is the identify the siRNA modifications that enable cytosolic delivery. However, the  proposal was not funded.,

A CEG pilot grant proposal has been successufully submitted and secured funding support  to investigate the role of UVR-induced tRNA modifications in melanoma tumor progression and metastasis.,
