Professional Summary

A long-term goal of my research program has been to gain an understanding of the structure and functioning of the cation transporter, Na+,K+-ATPase. Of particular interest is the elucidation of how cardiac glycosides (digitalis) bind and inhibit this enzyme’s function by altering its ability to undergo its normal conformational changes. We are also using anti-digitalis monoclonal antibodies, whose structures are known, as model drug receptors in order to better understand drug-receptor binding. These antibodies have also been used as binding templates to identify peptides that may mimic the properties of digitalis. Further, in collaborative efforts with Biotechnology companies we are developing novel, human sequence monoclonal antibodies directed against digitalis (digoxin) and cocaine for therapeutic use in patients. Our experimental approaches to these studies emphasize the use of poly and monoclonal antibodies, combinatorial peptide libraries, immunochemical, kinetic and fluorescence spectroscopy techniques as research tools. Currently, we are determining the effects of our anti-cocaine mAb on the in vivo properties of cocaine and utilizing computer-aided structural-modeling techniques to further aid our studies of drug-receptor/antibody interactions and the determination of enzyme and antibody structures.
