Research and Practice Interests

My lab is interested in studying the interplay between adipose tissue and bone.  The skeleton is traditionally thought of as a structural scaffold to support body weight and as a store for calcium.  However more recently its been shown to have a role metabolism and energy homeostasis.  Bone marrow adipose tissue (BMAT), which share a common progenitor as osteoblasts, has a complex relationship with bone.   BMAT is increases in osteoporosis, cancer, aging, and paradoxically in calorie restriction, notably also all conditions that are associated with lower bone mass.  Similarly, chronic corticosteroid exposure is well known to cause decline in bone density and increase in adiposity.   In one specific project we are investigating the role of dexras1 in this process.  Dexras1 gene expression is highly upregulated by corticosteroids and has been previously reported to promote adipogenesis in white adipose tissue.  We have found that it similarly promotes adipogenesis in bone marrow derived mesemchymal stem cells, and that its knockout in mice result in increased cortical and trabecular bone density.  It does not appear to have any obvious impact on the HPA axis.  We therefore hypothesize that dexras1 may be a mediator of adverse effects of corticosteroids, including obesity and osteoporosis.  If confirmed, it would be an attractive target for the treatment or preventions of corticosteroid related adverse effects.
