Susan Kasper, PhD, is Professor of Environmental Health at the University of Cincinnati, College of Medicine. Dr. Kasper received her PhD in Physiology/Endocrinology in the Laboratory of Henry G. Friesen, MD at the University of Manitoba, Winnipeg, Manitoba, Canada and completed a Medical Research Council (MRC) of Canada-funded postdoctoral fellowship in the Dana Farber Cancer Institute, Harvard Medical School and the Department of Molecular Medicine, New England Medical Center Hospitals, Boston, MA. Dr. Kasper joined the Department of Urologic Surgery at the Vanderbilt University Medical Center, Nashville, TN as Research Assistant Professor in 1996, moving to Assistant Professor (tenure track) and subsequently to Associate Professor (tenured) and Professor in the Department of Environmental Health at the University of Cincinnati.
 
Dr. Kasper has developed and currently leads a research program centered on processes that promote prostate cancer development and progression, including androgen receptor signaling, EMT, and signaling pathways which regulate self-renewal, proliferation, survival, adhesion, migration and differentiation of cancer stem cells (CSCs). These studies have received funding from the Department of Defense Prostate Cancer Research Program and NIH/NIDDK. New areas of research include the role of flame retardants as endocrine disruptors (in collaboration with Dr. Adam Biales, Chief at the Internal Exposure Indicators Branch (IEIB), National Exposure Research Laboratory, EPA at Cincinnati) and the role of exosomes in carrying signals that promote cancer/cancer progression (prostate cancer; head and neck cancers (in collaboration with Scott Langevin, PhD).

Research and Practice Interests

Research in our laboratory focuses on mechanisms by which prostate cancer becomes metastatic and progresses to therapeutic resistance. Therefore we are interrogating the integration of intrinsic (hormone receptor) and extrinsic (stem cell niche factor, environmental toxicant) signaling pathways in regulating genetic programs that control self-renewal, proliferation, survival, cell adhesion, migration and differentiation of stem/cancer stem cells. We have established several cancer stem cell-like models, including the human prostate HPET and HuSLC cell lines, which are being used to study these processes in vitro and in vivo (mouse xenograft model).
 
A second area of research focuses on Stathmin (Stmn1), a phospho-protein overexpressed in most cancers. Increased Stmn1 expression correlates with disease progression and poor prognostic outcome and is being considered as a potential therapeutic target. Stmn1 activity is tightly regulated by the differential phosphorylation of 4 serines in response to several major signaling pathways. Our study investigates the role(s) of these serines in regulating cell cycle progression, microtubule formation and epithelial mesenchymal transition (EMT) in vitro using a CRISPR approach to generate DU-145 prostate cancer cell lines carrying substitution mutations at these sites alone and in combination, and in vivo using an innovative zebrafish xenograft model. Knowledge on which Stmn1 phospho-forms control PCa cell proliferation and/or metastatic potential in the continuum of PCa progression is critical for the development of rational approaches to treat advanced/metastatic disease.
 
Recently, we have initiated studies in collaboration with Dr. Adam Biales, Chief at the Internal Exposure Indicators Branch (IEIB), National Exposure Research Laboratory, EPA at Cincinnati on the endocrine disrupting activities of Firemaster® 550 (FM550), a new-generation flame-retardant mixture that has replaced TDCPP in polyurethane foam. Trace levels of flame retardants are detected in multiple tissues, including adipose, liver, muscle, skin and blood in most Americans, when tested. Moreover specific populations are at much greater risk of exposure to high levels of flame retardants. An example is firefighters who have been diagnosed with PCa as early as in their 40s and 50s, as compared to the general male population who are on average > 65 years old at the time of diagnosis. Therefore there is a significant need to investigate the endocrine disrupting activities of flame retardants and determine their long-range effects on human health.
