Research and Practice Interests

My work is presently focused on the biochemistry and molecular biology of intracellular parasitism with particular reference to Leishmania donovani, a blood parasite of humans and domestic animals. These parasites have a life cycle comprising two stages: the flagellated promastigote found in the insect vectors and the nonflagellated amastigote found within the macrophages of the vertebrate host. Amastigotes thrive in the acidic environment of the phagolysosomes in macrophages where most other microorganisms cannot survive. A proton pumping plasma membrane ATPase has been identified as a key enzyme for intracellular survival of the parasite. It is pivotal in the regulation of intracellular pH within the acidic environment and in establishing a proton gradient across the membrane which drives active transport of nutrients. Using a combination of biochemical and molecular cloning techniques, the structure, function and regulation of this ATPase are being investigated.
