Professional Summary
Nancy Ratner, PhD, is interested in understanding mechanisms of peripheral nerve tumor (neurofibroma) formation in neurofibromatosis type 1 (NF1), a common inherited disorder in which children are predisposed to cancer of the nervous system, to learning problems, bone disorders, and other cancers. She identified EGFR and MEK as potential therapeutic targets in NF1 peripheral nerve tumorigenesis, and has developed cell culture and mouse models of NF1 nerve tumorigenesis. Her laboratory has also used analysis of gene expression to identify critical genes in neurofibroma and their malignant derivatives, MPNST.

Research and Practice Interests
Neurofibromatosis type 1, NF-1, is a common inherited disease of humans. Patients with NF-1develop a variety of symptoms, the most common of which are peripheral nerve tumors made up largely of Schwann cells, fibroblasts, and extracellular matrix. Neurofibromas develop sporadically in NF-1 patients, suggesting that mutations in the NF-1 gene cause only a predisposition to formation of tumors, which are then triggered by additional, or ''secondary'' genetic or epigenetic events. We have defined cellular abnormalities in neurofibroma cells. To understand the sequence of events which leads to neurofibroma formation, we test whether each of these abnormalities is a direct consequence of the NF-1 mutation. Experiments in progress use mice with targeted NF1 mutations, and primary cells from human tumors. These experiments will identify steps in formation of neurofibromas which could be useful in the development of clinical treatment of NF-1. Furthermore, as the NF1 gene product is a GTPase activating protein toward Ras, we are intersted in the relevance of blocking Ras activation as a method for reversing cellular phenotypes in cells that lack NF1, and ultimately for treating human NF1 patients.
