Extracellular signals generated by diverse stimuli, such as growth factors and environmental chemicals, trigger a cascade of events from the cell membrane to the nucleus. Our lab is interested in understanding how these signals are coordinated and integrated to evoke a distinct cellular response such as growth, proliferation, differentiation or death. We focus on the interplay of a variety of signaling pathways to understand the regulation of cellular signals and biological outcomes.
Our current research is focused on the MAP3K1 signaling pathway. MAP3K1 is a protein kinase that regulates cascades of signaling events in response to the growth and morphogenetic factors. This signaling pathway is critical for eyelid morphogenesis in fetal development. Our research objective is to determine how the MAP3K1 pathway integrates diverse growth and stress signals and regulates tissue development in the biological systems. Since MAP3K1 regulates the most fundamental biological activity, i.e., epithelial cell migration, it can be implicated in a variety of migration-related processes, such as embryonic development, tumor metastasis and injury repair. We also aim to understand how deregulation of the MAP3K1 pathway through gene-gene and gene-environment interactions can lead to diseases, such as birth defect and cancer progression. Our research should provide insights into the migratory- and morphogenetic-regulatory mechanisms, identify causative agents, and lead to prevention and treatment of diseases.
MAP3K1 is a tissue- and stimuli-specific regulator of the MAP2K-MAPK signaling cascades. MAP3K1 is temporal-spatially expressed in developing embryo/fetus and adult tissues; its activity is induced by a wide variety of stimuli. Being a player in the network of physiological and environmental factor-elicited signaling pathways, MAP3K1 is responsible for activation of the JNK-c-Jun pathway, leading to gene expression, cytoskeleton organization, cell migration and eyelid morphogenesis. Our research goal is to understand the mechanisms of MAP3K1 signaling focusing on elucidating how MAP3K1 expression and activity is regulated and what are the ways through which MAP3K1 integrates multiple signals to regulate cell migration and morphogenesis. Our studies will address the roles of MAP3K1 in signal transduction and biological processes using a combination of genetic, biochemical, cellular and molecular biological techniques and investigate the mechanisms of gene-gene and gene-environment interaction in organisms’ homeostasis and diseases using genetic mouse models.

Research and Practice Interests

The research focuses on establishing an understanding of the role of signal transduction pathways in cell responses to physiological and environmental agents. The ongoing work employs genetic, biochemical and molecular biological techniques to address fundamental questions regarding the function and regulatory mechanisms of signaling kinases in the areas of environmental toxicology, developmental biology, tissue regeneration, and cancer prevention and treatment.
