I. Research Interests
Gastric cancer develops from long-term inflammation that is typically caused by the bacteria Helicobacter pylori (H. pylori). During H. pylori-induced inflammation to cancer there are a number of changes that occur within the gastric epithelium. These include loss of gastric epithelial cell differentiation, parietal cell atrophy and infiltration of lymphocytes. Although it is accepted that inflammation precedes these pre-neoplastic changes, the mechanism by which inflammation triggers the development of cancer is unknown. Sonic Hedgehog (Shh), a morphogen originally identified for its role in embryogenesis, is believed to regulate adult epithelial cell homeostasis. In fact, coincident with H. pylori infection is the loss of Shh expression and this would then be expected to result in loss of normal epithelial cell differentiation. The overall focus and objective of my research is to define the mechanism(s) by which Shh acts as a constituent of gastric epithelial cell homeostasis and carcinogenesis. The detailed topics of my research projects are as follows:
Project 1: What is the mechanism by which Hedgehog signaling acts as a regulator of gastric epithelial cell differentiation and function?Project 1 investigates the regulation of normal differentiation and function of the adult stomach as impacted by the cellular location of Shh expression and secretion within the stomach to. Data from our laboratory has shown that the Hedgehog signaling pathway regulates the expression of E cadherin, and the loss of Hedgehog signaling in the stomach subsequently results in disruption of E cadherin expression and translocation of beta catenin from the cell membrane to the cytoplasm and nucleus. This results in the activation the Wnt pathway, increased proliferation, disruption of normal gastric epithelial differentiation and function.
Project 2: What is the mechanism by which Sonic Hedgehog regulates the host immune response during H. pylori infection?
The H. pylori infected mucosa exhibits a T-helper 1 (TH1) immune response characterized by recruitment of primarily interferon gamma (IFNg)-expressing T lymphocytes to the stomach. Although gastritis due to H. pylori is induced by a TH1-mediated response that is T cell and IFNgdependent, the mechanism regulating the development of such an inflammatory response is unclear. In addition to the essential role for patterning during embryogenesis and maintenance of adult tissue homeostasis, Shh is also known to be a regulator of T cell differentiation and cytokine production. However, the role of Shh in the development of H. pylori-induced gastritis has not been investigated. Project 2 studies the role of Hedgehog signaling in the development of the initial host immune response triggered by bacterial infection.
Project 3: What is the role of Hedgehog signaling in the development of gastric cancer?
In gastric cancer tissue, histological alterations and changes in cell composition coincide with the over-expression of Shh that contributes to angiogenesis and tumor growth.
